The LysoSensor® series of probes are fluorescent probes used to detect the pH value of acidic compartments in living cells (such as lysosomes and acrosomes). These probes exhibit acidophilic staining and accumulate in acidic organelles through protonation. This acidophilicity of the probes overcomes the fluorescence quenching effect of the dye, resulting in stronger fluorescence emission. Therefore, compared to the LysoTracker series of probes, the fluorescence intensity of the LysoSensor® series of probes increases in a pH-dependent manner (towards acidity) with the degree of organelle acidification.
This product, LysoSensor™ Yellow/Blue DND-160 (PDMPO), is a ratiometric probe for measuring lysosomal pH. It is a pH-dependent fluorescent probe in the LysoTracker® series, displaying dual excitation and dual emission wavelengths. This probe produces blue fluorescence in a neutral environment (Ex/Em = 329 nm/440 nm), but emits yellow fluorescence (Ex/Em = 384 nm/540 nm) when the environment becomes more acidic. This product is supplied as a 1 mM stock solution dissolved in anhydrous DMSO.

体内配方配制方法:取 μL DMSO母液,加入 μL PEG300,混匀澄清后加入μL Tween 80,混匀澄清后加入 μL saline,混匀澄清。

一定要按照顺序依次将溶剂加入,进行下一步操作之前必须保证上一步操作得到的是澄清的溶液,可采用涡旋、超声或水浴加热等物理方法助溶。
