Heparinase II is a glycosidase targeting heparin and heparan sulfate. Heparinase II is promising for research of glycosaminoglycan structure-function analysis, anticoagulant agents and tumor microenvironment modulation[1].
Product Information
Calcium ions inhibit heparinase II activity. Heparinase II cleaves sulfated polysaccharide chains containing 1-4 linkages at the junctions of hexosamine and uronic acid residues (including iduronic acid and glucuronic acid residues) via an elimination mechanism. This reaction produces oligosaccharide products (mainly disaccharides) containing unsaturated uronic acids, detectable by UV spectroscopy at 232 nm. The enzyme also cleaves the antithrombin III-binding pentasaccharide domain in the heparin molecule.
Dissolution
Use 1 mg/mL dissolved in water; fresh preparation is recommended. Alternatively, dissolve according to the specific experimental guidelines.
Reaction Conditions
1. Stable pH range: 5.0-9.0, optimal pH: 7.0-7.5
2. Stable temperature range: 20-37℃, optimal temperature: 30℃
Note: Incubation time and enzyme concentration need to be determined based on practical experience when digesting the substrate.
