TRFS-green is a turn-on green fluorescent probe with high selectivity for thioredoxin reductase (TrxR). TRFS-green is widely used for imaging TrxR activity in live cells, with Ex/Em of 377nm/480nm, and 438nm/538nm under simulated physiological conditions[1]. TrxR is often overexpressed in many cancer cells, and its ablation can reverse the growth of various malignancies, reinforcing this selenoenzyme as a valid target for the development of novel anticancer drugs[2,3,4]. TRFS-green can facilitate the discovery of TrxR-targeting small molecules and advance the understanding of the physiological and pathological functions of TrxR in vivo[1,5].

实验参考方法 Experimental Reference Method

TRFS-green的储存:

(1)室温离心TRFS-green至管底。

(2)用DMSO溶解,使TRFS-green终浓度为10mM,并分装冻存,避免反复冻融。

使用TRFS-green进行活细胞成像[1]

(1)将Hep G2细胞接种于12孔板中,并在补充了10%FBS、2mM glutamine、penicillin(100units/mL)、streptomycin(100units/mL)的DMEM培养基中,于37℃、5%CO2的湿润环境下过夜培养。

(2)加入TRFS-green(10μM),并将细胞继续孵育2-4h。

(3)用培养基冲洗细胞3次以去除残留的TRFS-green。

(4)使用荧光显微镜获取荧光图像,使用荧光微孔板读数仪对细胞荧光强度进行定量(Ex/Em: 438nm/538nm)。
