N-methyl Mesoporphyrin IX (NMM) is a transition state analog of porphyrin and an effective inhibitor of ferrochelatase with a Kd value of 7nM[1]. N-methyl Mesoporphyrin IX is a specific stain for G-quadruplexes, with excitation/emission maxima at 399/610 nm[2, 3]. G-quadruplexes (G4s) are higher-order structures formed by DNA or RNA sequences rich in tandem guanine repeats, and are stabilized by G-quartets through π-π stacking[4]. N-methyl Mesoporphyrin IX can serve as an effective probe for monitoring Aβ-amyloid aggregation in live cells related to Alzheimer's disease[5]. N-methyl Mesoporphyrin IX can stabilize human telomeric DNA dAG3 (TTAG3)3 (Tel22) and selectively bind to its parallel conformation with a 1:1 stoichiometry and a binding constant of approximately 1.0 × 105 M−1[6].

实验参考方法 Experimental Reference Method

本方案仅提供一个指导,请根据您的具体需要进行修改。

1. 溶液配制
(1)储存液:用DMSO溶解N-methyl Mesoporphyrin IX,终浓度为5-10mg/mL,可根据实际需要对储存液浓度进行调整。
注意:未使用的储存液分装后在-20℃避光保存,避免反复冻融。

(2)工作液:用实验缓冲液稀释储存液到所需的工作浓度。
注意:最佳的工作浓度请根据实际情况调整或参阅文献自行设置梯度浓度进行摸索。工作液必须现配现用。保证DMSO含量低于千分之一(1‰)。

2.使用N-methyl Mesoporphyrin IX对凝胶内G-四链体染色的实验步骤[1](来自文献,仅做参考)
(1)将200 pmol的非标记寡聚物在20μL 50mM KCl中折叠。将折叠的寡聚物与20μL上样缓冲液混合。取20μL混合样品加载到含有5%甘油的12%聚丙烯酰胺凝胶上。
(2)进行电泳实验,电泳结束后将凝胶取下用于染色
(3)准备N-methyl Mesoporphyrin IX储备液(5 mg/mL),溶于0.2 M HCl。
(4)准备N-methyl Mesoporphyrin IX工作液(1 μg/mL),用20 mM Tris(pH 7.6)、100 mM KCl、1 mM EDTA稀释。浸没凝胶孵育20分钟。
(5)使用荧光图像分析仪在532nm波长下扫描凝胶。

3.使用N-methyl Mesoporphyrin IX测定Aβ40的实验步骤[2](来自文献,仅做参考)
(1)作为离体探针(ex situ probe):将N-methyl Mesoporphyrin IX加入已孵育的Aβ40溶液中,浓度为1 μM。
(2)作为原位抑制剂(in situ inhibitor):将N-methyl Mesoporphyrin IX(10 μM)与Aβ40(50 μM)在37°C下共孵育7天。在荧光测量前加入额外的N-methyl Mesoporphyrin IX,使浓度保持在1 μM。
(3)使用荧光图像分析仪在激发波长399 nm下,收集550到700 nm范围内N-methyl Mesoporphyrin IX的光谱数据。
