GGTI 286 is an inhibitor of geranylgeranyltransferase I (GGTase I) that selectively inhibits geranylgeranylation of Rap1A over farnesylation of H-Ras in NIH3T3 cells (IC50s = 2 and >30 μM, respectively). It also inhibits geranylgeranylation of oncogenic K-Ras4B in NIH3T3 cells (IC50 = 2 μM) and blocks K-Ras4B-induced activation of MAP kinase (IC50 = 1 μM). GGTI 286 (2.5-50 μM) is cytotoxic to RPMI-8226, H929, and U266 multiple myeloma cells in a concentration-dependent manner. It also reduces angiotensin II-induced collagen mRNA and protein production in rat cardiac fibroblasts.
