DAF-FM DA (Diaminofluorescein-FM diacetate) is a cell-permeable fluorescent probe used for the detection and bioimaging of nitric oxide (NO). The DAF-FM DA (Diaminofluorescein-FM diacetate) probe can quantify the level of nitric oxide (NO) in cells, with a maximum excitation/emission wavelength of 495/515nm[1-2].
DAF-FM DA (Diaminofluorescein-FM diacetate) can permeate the cell membrane of living cells and is deacetylated by intracellular esterases to become DAF-FM. DAF-FM can react with NO produced in cells to form highly fluorescent triazole fluorescein (DAF-FMT), which can be located and monitored by fluorescence or confocal microscopy, or detected by fluorescence assay and flow cytometry. The NO detection limit of DAF-FM DA (Diaminofluorescein-FM diacetate) is approximately 3nM[1-3]. DAF-FM DA (Diaminofluorescein-FM diacetate) can be used as a vital dye for zebrafish embryos to mark the early ossification centers of developing zebrafish[4]. DAF-FM DA (Diaminofluorescein-FM diacetate) can be used to detect the production of NO in fungi[5].
