(Z-Ala-Ala-Ala-Ala)2Rh110 is a sensitive, non-fluorescent elastase substrate. Upon selective cleavage by elastase, the colorless (Z-Ala-Ala-Ala-Ala)2Rh110 yields the strongly fluorescent compound Rhodamine 110, which exhibits excitation and emission wavelengths of 485nm and 525nm, respectively[1, 2]. (Z-Ala-Ala-Ala-Ala)2Rh110 demonstrates high specificity for elastase with minimal cross-reactivity, making it suitable for the precise quantitative analysis of enzymatic activity within complex biological samples. For instance, (Z-Ala-Ala-Ala-Ala)2Rh110 can be utilized to determine the enzymatic activity of neutrophil elastase (NE) in the bronchoalveolar lavage fluid (BALF) of mice infected with the influenza A virus (IAV)[3, 4].
