Soyasaponin BB is an oleanane-type triterpenoid saponin derived from soybeans (a leguminous plant). As an orally active molecule, Soyasaponin BB exhibits a dual mechanism of action by inducing the expression of heme oxygenase-1 (HO-1) while simultaneously inhibiting the aldose reductase AKR1B1[1-2]. Soyasaponin BB helps reduce inflammatory responses and promotes cartilage tissue regeneration[3]. Soyasaponin BB can also alleviate LPS-induced cognitive dysfunction and intestinal inflammation[4].
In vitro, when C2C12 cells were co-cultured with Soyasaponin BB (10–30μg/mL) for 3 days under the induction of bone morphogenetic protein-2 (BMP-2; 100ng/mL), Soyasaponin BB promoted their differentiation into osteoblasts and significantly enhanced the activity and expression of alkaline phosphatase (ALP). Soyasaponin BB activates the AKT/MAPK signaling pathway, thereby promoting the transcription of downstream osteogenic markers such as osteocalcin (OCL) and type I collagen (Col1a)[5]. Pretreatment of BRL 3A rat hepatocytes with Soyasaponin BB (25–100μg/mL) for 24 hours, followed by stimulation with ethanol (250mmol/L) for another 24 hours, significantly alleviated ethanol-induced oxidative stress and hepatocyte damage[6].
In vivo, intragastric administration of Soyasaponin BB (10mg/kg; for 4 consecutive days) to male C57BL mice undergoing spontaneous withdrawal after chronic intermittent escalating-dose morphine administration significantly reduced withdrawal-induced anxiety-like behaviors[7]. Dietary supplementation with Soyasaponin BB (0.2g/100g feed) for 90 days in 60-day-old male pcy mice markedly delayed the progression of polycystic kidney disease[8].

在体内,Soyasaponin BB(10mg/kg;连续4天)通过胃内给药处理经历慢性间歇性递增剂量吗啡给药后自发戒断的雄性C57BL小鼠,Soyasaponin BB显著减轻了戒断诱导的焦虑样行为[7]。Soyasaponin BB(0.2g/100g饲料)通过膳食补充方式处理60日龄雄性pcy小鼠90天,显著延缓了多囊肾病的疾病进展[8]。
