(S)-Pro-xylane, a biologically active C-glycoside compound, is an important ingredient in antiaging skincare formulations in cosmetic products [1]. Through mimicking natural β-O-xylosides in skin, (S)-Pro-xylane is effective in stimulating the biosynthesis of glycosaminoglycans (GAGs), which leads to the preservation of matrix integrity and thereby contributes to skin's mechanical properties, such as elasticity and hydration [2]. (S)-Pro-xylane has been widely used to prevent the degradation and loss of glycosaminoglycans and proteoglycans in vivo[3].

In vitro, (S)-Pro-xylane treatment at 0.4mg/ml for 18h significantly enhanced the mRNA levels of collagen VII a1 and collagen IV a1 in normal human epidermal keratinocytes (NHKs), without affecting cell viability[4]. Treatment with 3mM (S)-Pro-xylane for 3 days increased syndecan 4 and perlecan levels in an ex vivo atrophic skin model[5].

In vivo, topical application of 100μl 1.5mg/ml (S)-Pro-xylane aqueous solution daily for 3 weeks significantly improved collagen loss and fiber breakage associated with skin aging, and reduced the degradation of elastic fibers associated with skin aging in Sprague-Dawley (SD) rats[6].

Preparation Method

Normal human dermal fibroblasts (NHDFs) were cultured in DMEM medium supplemented with 10% fetal bovine serum, 1% penicillin-streptomycin and amphotericin B triple antibiotic solution. Cells were cultured in a 5% CO2 incubator at 37°C. Cells were treated with 0.4mg/ml of (S)-Pro-xylane for 18h and analyzed for cell viability and mRNA levels of collagen VII a1 and collagen IV a1.

Applications

(S)-Pro-xylane treatment significantly enhanced the mRNA levels of collagen VII a1 and collagen IV a1, without affecting cell viability.

Animal models

Male Sprague-Dawley rats

Preparation Method

Male Sprague-Dawley rats were housed under controlled conditions (22±3°C, 12/12h light/dark cycle) and randomly assigned to experimental groups. Two skin areas, each measuring 2cm×2cm, were selected from the back of each rat, and the hair at the sampling sites was shaved. For each rat, one skin area was designated as the negative control, while the other was used as the (S)-Pro-xylane-treated group. The skin in the (S)-Pro-xylane-treated group was topically applied with 100μl 1.5mg/ml (S)-Pro-xylane aqueous solution (containing 10% propylene glycol) daily for 3 weeks. Rat skin morphology, collagen fibers, and elastic fibers were analyzed.

Applications

(S)-Pro-xylane treatment can improve the skin condition of rats, resulting in an increase in the content of collagen fibers and elastic fibers in the skin.
