1,6-Diphenyl-1,3,5-hexatriene is a fluorescent probe for lipid monolayers and bilayer membranes (with maximum excitation and emission wavelengths of 360/430nm). 1,6-Diphenyl-1,3,5-hexatriene can be localized in the hydrocarbon region of membrane lipids, exhibiting almost no fluorescence in water, but its fluorescence increases significantly upon membrane insertion, thereby enabling the detection of membrane viscosity, polarity, and lipid order. 1,6-Diphenyl-1,3,5-hexatriene can be used in studies of membrane fluidity, lipid droplet fluorescence detection, determination of surfactant critical micelle concentration, and reporting of bacterial spore membrane fluidity, among other related research[1-4].

1,6-Diphenyl-1,3,5-hexatriene荧光探针在膜研究中的使用方法[1]
本实验方案根据研究内容整理,仅供参考,请根据实际需求进行调整。
1. 试剂准备:将1,6-Diphenyl-1,3,5-hexatriene溶解于二甲基甲酰胺(DMF)中,配制成储备液;将POPC(34.6mM)和ITZ(9.5mM)分别溶解于氯仿中。
2. 脂质体制备(小单层脂质体SUVs):取适量POPC和ITZ氯仿储备液混合于容量瓶中,用氮气流吹干溶剂形成干燥脂膜。加入磷酸盐缓冲液(终浓度POPC为2.5mg/mL)水化,超声处理得到粒径30-200nm的SUVs。
3. 1,6-Diphenyl-1,3,5-hexatriene标记:向脂质体样品中加入DPH储备液,使1,6-Diphenyl-1,3,5-hexatriene终浓度为102nM,在黑暗条件下孵育1小时。
4. 稳态荧光各向异性测量:使用光谱荧光计,设置激发波长λ_exc=360nm,发射波长λ_em=430nm,测量垂直和水平偏振的荧光强度。

(1)1,6-Diphenyl-1,3,5-hexatriene荧光寿命对膜环境的极性(介电常数)敏感,寿命值在更有序、更疏水的膜环境中会延长。
