Tos-Gly-Pro-Arg-AMC . HCl is a fluorescent substrate for thrombin, which is widely used to detect serine protease activity. Its excitation and emission wavelengths are 380nm and 460nm, respectively.

The core structure of Tos-Gly-Pro-Arg-AMC . HCl contains a fluorescent group 7-amino-4-methylcoumarin (AMC). When the substrate is hydrolyzed by protease, AMC is released, generating a strong fluorescent signal (excitation wavelength 380nm, emission wavelength 460nm). The Gly-Pro-Arg sequence is a specific recognition site for thrombin, so this substrate is highly selective for thrombin and is also suitable for the study of other serine proteases (such as trypsin, kallikrein, etc.). Tos-Gly-Pro-Arg-AMC . HCl can also be used for high-throughput screening of thrombin inhibitors or for detecting thrombin activity in blood samples to assist in the diagnosis of coagulation abnormalities[1][2].

结肠上清液中的胰蛋白酶样活性检测[1][2]
1. 溶液准备
底物溶液:将Tos-Gly-Pro-Arg-AMC . HCl溶解在50mM Tris-HCl(pH 8.0)缓冲液中,配制成10mM的储存液。使用前稀释至工作浓度100µM,并预热至室温。
2. 样品制备
A: 取50mg远端结肠样本,用Krebs溶液冲洗后速冻并存于-80℃。
B: 使用液氮粉碎样本以确保酶活性,加入蛋白裂解缓冲液裂解15min,离心后收集上清液备用。
3. 荧光检测
A:将结肠上清液置于冷96孔板中,确保样品保持低温以避免酶活性损失。
B:加入预热至室温的底物溶液,孵育20min。
C:使用酶标仪在激发波长380nm、发射波长460nm的条件下动态检测AMC的荧光强度变化,并计算酶活性。
