Z-Gly-Gly-Arg-AMC is a specific thrombin fluorescent substrate with λexc = 355nm, λem = 460nm[1]. Z-Gly-Gly-Arg-AMC acts as the reference substrate in the Calibrated Automated Thrombogram (CAT) for measuring endogenous thrombin potential (ETP) in platelet-rich (PRP) or platelet-poor plasma (PPP)[2]. Z-Gly-Gly-Arg-AMC is employed in 96-well format for rapid identification and potency ranking of direct thrombin inhibitors or protease-activated receptor antagonists[3].

基于Z-Gly-Gly-Arg-AMC检测凝血酶活性[1]

本方案仅提供了一种指导原则,具体操作应根据您的实际需求进行调整。

1.溶液配制

(1)将Z-Gly-Gly-Arg-AMC固体溶解在适量的DMSO中,以配制50mM的储备溶液。将储备溶液存放在-20°C低温环境中保存,并避免反复冻融。

(2)取适量的Z-Gly-Gly-Arg-AMC储备溶液在PBS中稀释,以配制2.5mM的工作溶液。工作溶液应现配现用。

(3)取适量的CaCl2溶液,并溶解在检测缓冲液(含有20mM的HEPES、145mM的NaCl,pH 7.5)中,以配制激活缓冲液。

2. 凝血酶生成测定

(1)将血浆与Z-Gly-Gly-Arg-AMC储备溶液按体积比 4:1 混合,将混合液置于平底96孔板的孔中。向每个孔中加入100µl混合液,然后向每个孔中加入20µl激活缓冲液以触发凝血反应。Z-Gly-Gly-Arg-AMC的最终浓度为400µM。

(2)使用荧光板读数仪(Ex/Em=355/460nm)在37°C下连续测量荧光,持续30分钟,每3分钟读取一次。

3. 注意事项

(1)Z-Gly-Gly-Arg-AMC具有光敏性,操作过程应避光进行。

(2)为避免产生气泡,应在添加激活缓冲液(含CaCl2)后的10秒内进行读数。
