TAMRA azide, 5- isomer is a fluorescent probe that contains an azide group, with the spectral characteristics (excitation 562/40nm, emission 624/40nm)[1]. TAMRA azide, 5-isomer is highly suitable for different click reactions or the Staudinger ligation, and reacting with alkyne, DBCO and BCN[2].

TAMRA azide was used as a fluorophore to label histone H3 and allowed efficient labeling of the modified substrate by CuAAC click chemistry[3]. TAMRA azide has been used for protein microarray immobilization to develop protein biosensors[2]. In HEK293T cells, TAMRA azide has been used in click chemistry to detect S-palmitoylation[4].

实验参考方法 Experimental Reference Method

TAMRA azide可通过点击化学反应用于炔烃标记物Alk14(炔14)的信号检测:
转染质粒并用50μM Alk14处理HEK293T细胞6小时后,分别使用抗Flag M2树脂和GFP-Trap对Flag-NLRP3及NLRP3-GFP进行免疫共沉淀。
琼脂糖珠经免疫沉淀洗涤缓冲液清洗三次后,重悬于20μL相同缓冲液中。
向珠悬液中加入点击化学反应混合试剂(含2mM TAMRA azide、10mM TBTA、40mM CuSO4、40mM TCEP,各组分1μL),避光室温孵育。
30分钟后加入10μL SDS上样缓冲液终止反应,95℃煮沸10分钟。
煮沸后的样品离心取上清,转移至新管中进行羟胺处理(终浓度0.5M),再次煮沸5分钟。
最后通过SDS-PAGE电泳和ChemiDoc成像系统进行凝胶内荧光分析,检测Alk14标记信号。
