Sulfatase from Helix pomatia is a highly efficient enzyme from Helix pomatia that hydrolyzes sulfated compounds and sulfate esters[1]. Sulfatase from Helix pomatia can be used to extract lignans from oilseed mixtures by degrading glycosidic bonds and releasing lignan aglycones[2]. Sulfatase from Helix pomatia can be used for glucosinolate analysis and genistein extraction preparation[3, 4].

1. 溶液配制

(1)工作液:用0.05M乙酸钠缓冲液溶解Sulfatase from Helix pomatia,配制浓度为2mg/mL的工作液。

注意:最佳的工作浓度请根据实际情况调整或参阅文献自行设置梯度浓度进行摸索。工作液必须现配现用。

2. 使用Sulfatase from Helix pomatia进行木脂素提取[1](来源文献,仅供参考)

(1)用0.5ml 100%甲醇提取1mL发酵样品,超声处理10min,然后轻轻搅拌10min。

(2)在4°C下以13200rpm的速度离心样品10min,将上清液收集到2mL干净的EP管中,并在氮气流下蒸发干燥。

(3)加入0.6mL Sulfatase from Helix pomatia,置于37°C下孵育过夜并轻轻搅拌。

(4)样品冷却,加入0.5mL 酸化水(0.4%甲酸),涡旋,然后在4°C下以 13200rpm离心10min。将上清液收集到干净的EP管中进行固相萃取。
