实验参考方法 Experimental Reference Method

点击棕榈酰化试验

1.HEK293T细胞铺板,用质粒(YKT6-WT BioID、YKT6-3E BioID和Wnt3A)转染添加10%胎牛血清的DMEM中。

2.ω-炔基棕榈酸(Palmitic Acid Alkyne)在乙醇中溶解至终浓度为50mM,保存于- 80°C。

3.将Palmitic acid alkyne在添加5% FBS(不含脂肪酸)的DMEM中稀释至终浓度为100μM,在室温水浴中超声15min,然后再进行预络合15min。

4.加入含Palmitic Acid Alkyne的培养基,24h后部分更换。

5.转染后72h,裂解细胞(含1% Triton x-100的PBS, 0.1% SDS, PIC),然后在4℃下,16000 g离心5min。

6.用Wessel-Flugge蛋白沉淀法沉淀裂解物。

7.将点击标记反应物[0.1mM叠氮生物素,1mM盐酸三(2-羧乙基)膦]溶于水中

8.与溶解于DMSO中的0.1mM Tris[(1-苄基- 1h -1,2,3-三唑-4-酰基)甲基]胺和溶解于水中的1mM CuSO4在37℃黑暗条件下混合摇动培养2h。

9.点击反应后,沉淀样品用10倍甲醇在- 80℃下过夜,然后离心,再次用冰冷的甲醇洗涤

10.将干燥后的颗粒在4% SDS中重悬。

11.用高容量中性琼脂糖树脂沉淀click-生物素化蛋白。

12.用1% SDS洗涤样品并洗脱,然后通过免疫印迹进一步分析。
