L-Ascorbic Acid 2-phosphate (magnesium salt hydrate) is a vitamin C derivative usually employed as a raw material in cell and tissue therapy[1]. L-Ascorbic Acid 2-phosphate is commonly used as a substrate for alkaline phosphatase assay[2].
In vitro, L-Ascorbic Acid 2-phosphate treatment (1000μM) for 3 weeks significantly stimulated the proliferation of human skin fibroblasts and induced collagen synthesis[3]. L-Ascorbic Acid 2-phosphate treatment (300μM) for 2 weeks significantly stimulated the growth of human corneal endothelial cells, maintained the characteristic polygonal morphology, reduced the generation of 8-hydroxy-2-deoxyguanosine (8-OHdG), and prevented oxidative DNA damage[4]. After 30 days of culture in the presence of L-Ascorbic Acid 2-phosphate (100μM), the growth of rabbit corneal cells was significantly increased, and the cells had multilayer characteristics[5]. L-Ascorbic Acid 2-phosphate (250μM) treatment for 7 days significantly inhibited the growth of adipose-derived stem cells seeded at low density (1,250 cells/cm2), resulting in decreased expression of antioxidant enzymes[6]. L-Ascorbic Acid 2-phosphate treatment (50μM; 24h) significantly and sustainably improved intracellular L-ascorbic acid and enhanced type 1 collagen synthesis and mRNA expression in human gingival fibroblasts, with reduction of intracellular reactive oxygen species (ROS)[7].
