Hydrogenated soybean phospholipids, commonly used as a liposomal carrier for drug delivery, can enhance the stability of liposomes composed of stratum corneum lipids (SC lipids) under neutral pH conditions[1-3]. Hydrogenated soybean phospholipids can enhance the encapsulation of curcumin in liposomes, improve the stability of curcumin-loaded liposomes, slow the release of curcumin during simulated digestion, and promote tighter lipid packing in liposomal membranes[4].

使用薄膜水化法制备载药雷公藤内酯醇的脂质体(LRA)药物递送系统[1]
1. Hydrogenated soybean phospholipids、胆固醇(CHO)、聚乙二醇化磷脂乙醇胺(DSPE-PEG2k)和雷公藤内酯醇(RA)按照3.3:2.3:0.3:1的摩尔比溶解于甲醇中。
2. 随后,使用旋转蒸发仪去除溶剂,得到脂质薄膜。
3. 通过使用去离子水水化干燥的脂质薄膜并搅拌120分钟,然后超声处理(功率500瓦)120分钟来制备脂质体。
4. 搅拌12小时后,通过透析(透析袋,截留分子量3500道尔顿)去除游离的RA。
5. 透析液通过0.45微米滤膜过滤,并加入2%(质量体积分数)的海藻糖进行冷冻干燥,最终得到LRA。
6. 通过高效液相色谱法(HPLC)确认了药物载药量(DLC%)。
7. DLC%的计算公式如下:DLC%=(RA的质量)/(总雷公藤内酯醇脂质体的质量)×100%。
8. 高效液相色谱法(HPLC)的条件如下:流动相为乙腈和水(含0.05%醋酸),流速为1毫升/分钟,乙腈与水的比例为65:35。
9. 使用紫外(UV)检测器,吸收波长为256纳米,用于测定RA的含量。
