Ethyl glucuronide is a nonvolatile, water-soluble, direct metabolite of ethanol, which can be detected in body fluids, tissues, and hair [1]. Ethyl glucuronide content is positively correlated with alcohol intake and can be used as a marker for the detection of alcohol abuse[2]. Ethyl glucuronide can be used as a standard internal control to develop a liquid chromatography-tandem mass spectrometry method for the identification of related compounds[3].
In vitro, Ethyl glucuronide treatment (100μM) for 24h blocked the formation of TLR4-MD2 complex and stimulated the expression of TLR4-dependent SEAP protein in HEK-TLR4 cells[4].
In vivo, Ethyl glucuronide treatment via intrathecal injection at a single dose of 0.38µg caused significant hyperalgesia in rats within 3 hours[4].

在体外,使用100µM的Ethyl glucuronide处理24小时,阻断了HEK-TLR4细胞中TLR4-MD2复合物的形成,并刺激了TLR4依赖性SEAP蛋白的表达[4]。
在体内,单次鞘内注射0.38µg剂量的Ethyl glucuronide,在3小时内导致大鼠出现显著的痛觉过敏[4]。

Preparation Method | HEK-TLR4 cells were cultured in DMEM medium supplemented with 10% fetal bovine serum (FBS) and antibiotics (penicillin 1000U/ml, streptomycin 10mg/ml) under normal conditions (5% CO2 with 95% humidified air). HEK-TLR4 cells (5×103 per well) were inoculated in a 96-well cell culture plate and incubated at 37°C with 5% CO2 for 48h. Then, the cells were treated with different concentrations of Ethyl glucuronide (0.01, 0.1, 1, 10, 100µM) for 48h, respectively, and the supernatant (15μl) was then collected from each well for measurement of SEAP levels. |
