Description

Pro-xylane is a bioactive C-glycoside derived from Fagus sylvatica xylose. Pro-xylane stimulates the synthesis of glycosaminoglycans (GAGs) and hyaluronic acid to fill the extracellular matrix and lock in moisture, while strengthening the dermo-epidermal junction by promoting the synthesis of type IV and VII collagens. Pro-xylane can be used in studies related to skin anti-aging and chronic wound healing[1-4].

In vitro, epidermal keratinocytes and dermal fibroblasts were treated with Pro-xylane (1–3mM) for 48 hours. Pro-xylane significantly induced filaggrin expression and promoted α-smooth muscle actin generation, while inhibiting the secretion of the inflammatory factor prostaglandin E2[5]. Pro-xylane (0.3–3mM) was applied to dermal fibroblasts for 72 hours. Pro-xylane significantly stimulated the synthesis of sulfated glycosaminoglycans (GAGs)[6].

Pro-xylane是一种源自山毛榉木糖的、具有生物活性的C-糖苷。Pro-xylane可刺激糖胺聚糖(GAGs)和透明质酸的合成来填充细胞外基质并锁水保湿,同时通过促进IV型和VII型胶原蛋白合成以强化真皮-表皮连接。Pro-xylane 可用于皮肤抗衰老和慢性伤口愈合的相关研究[1-4]

在体外,Pro-xylane(1–3mM)处理表皮角质形成细胞及真皮成纤维细胞48小时。Pro-xylane显著诱导丝聚蛋白表达并促进α-平滑肌肌动蛋白生成,同时抑制炎症因子前列腺素E2的分泌[5]。Pro-xylane(0.3–3mM)处理真皮成纤维细胞72小时。Pro-xylane显著刺激硫酸化糖胺聚糖(GAGs)的合成[6]

Human dermal fibroblasts (normal human dermal fibroblast, NHDF)

Preparation Method

Normal human dermal fibroblast (NHDF) monolayers were cultured in control medium with or without Pro-xylane at the indicated concentrations for 72h at 37°C in humid atmosphere of 95% air and 5% CO₂, and labeled with ³⁵S-sulfur during the final 24h.

Reaction Conditions

0.3mM-3.0mM; 72h

Applications

Pro-xylane significantly stimulated the biosynthesis of sulfated glycosaminoglycans (GAGs) by human fibroblasts in a dose-dependent manner, as evidenced by the increased incorporation of ³⁵S-radioactivity into the isolated GAG fraction.
