4-Methylumbelliferyl-β-D-Glucopyranoside is a β-D-glucoside that serves as a fluorescent substrate for β-glucosidase, ​thereby allowing for the detection and analysis of​ this enzyme's activity[1]. ​The enzymatic cleavage of​ 4-Methylumbelliferyl β-D-glucopyranoside ​results in the release of​ the highly fluorescent compound 4-Methylumbelliferone. The maximum emission wavelength of 4-MU ranges from 445 to 454nm, while its excitation wavelength varies with pH: at pH 4.6, 7.4, and 10.4, the corresponding excitation wavelengths are approximately 330nm, 370nm, and 385nm, respectively[2].

细胞中蛋白酶体活性测定
本实验方案根据研究内容整理,仅供参考,请根据实际需求进行调整。
1. 酶样品制备:将细胞裂解后,取上清液于4°C保存备用。
2. β-葡萄糖苷酶活性检测:反应体系包含5μM 4-Methylumbelliferyl-β-D-Glucopyranoside,100mM柠檬酸缓冲液(pH=5.0),总体积为200μL;45°C孵育1小时后,加入2.5mL 0.5M Na₂CO₃缓冲液(pH=10.4)终止反应。使用荧光光度计检测产物在激发波长350nm、发射波长440nm下检测产物。

1)游离的荧光底物存在淬灭问题,请尽量注意避光,以减缓荧光淬灭。
