Sulfo-NHS-LC-Biotin is a labeling reagent with a long-chain structure that reacts with amines. Its biotin-like groups can be efficiently eluted from streptomycin avidin, making it suitable for purification or labeling of cell surface proteins. The spacer arm between the biotin and amine reactive linker is large, aiding in overcoming steric hindrance and enhancing the efficiency of surface labeling [1-3].

Sulfo-NHS-LC-Biotin (10 mM) can be used for biotinization of exosomes from Mycobacterium tuberculosis (Mtb)-infected cells [4]. Sulfo-NHS-LC-Biotin also can be used for label cells[5]. It also can monitor sertoli cell Blood-Testis Barrier (BTB) integrity [6].

Sulfo-NHS-LC-Biotin (10 mM)可用于结核分枝杆菌(Mtb)感染细胞外泌体的生物素化[4]。Sulfo-NHS-LC-Biotin也可用于标记细胞[5]。它还可以监测sertoli细胞Blood-Testis Barrier (BTB)的完整性[6]。

实验参考方法 Experimental Reference Method

外泌体生物素化[1]：

Sulfo-NHS-LC-Biotin用于标记暴露于完整外泌体外部小叶的蛋白质，并且生物素和胺反应性连接剂之间含有较大的间隔臂，有助于克服位阻并提高外泌体表面的标记效率。

  1. 取200μg完整外泌体与10 mM Sulfo-NHS-LC-Biotin室温混合30 min。
  2. 孵育后，使用10 KDa MWCO过滤装置去除过量的Sulfo-NHS-LC-Biotin。
  3. 生物素化的外泌体用10倍体积的1×PBS洗涤，浓缩至终体积为30 μl。用300 ul RIPA缓冲液裂解生物素化外泌体1小时，然后进行3次冻融循环。
  4. 裂解后进行缓冲液交换，用1倍PBS代替RIPA缓冲液。

裂解的生物素化外泌体在室温下暴露于10 mM的Sulfo-NHS-Biotin中30分钟，以标记剩余的游离胺。最后去除过量的生物素。

细胞Blood-Testis Barrier (BTB)完整性监测[2]:

注:在PBS/CM pH 8.0 (10 mM磷酸钠，0.15 M NaCl)中新鲜配制10 mg/ml Sulfo-NHS-LC-Biotin，其中含有1 mM CaCl2作为标记缓冲液(生物素试剂在使用前新鲜配制;不要储存)。

  1. 将支持细胞接种于12孔培养板中，用3mL DMEM/F-12培养基培养4-5天，建立功能通透屏障。每48小时更换一次培养基，并补充生长因子。
  2. 用冷PBS (pH 7.4)清洗细胞，并将细胞清洗三次，以去除细胞中含胺的培养基和蛋白质。
  3. 每mL DMEM/F-12加入20 μL生物素试剂作为生物素化反应混合物(见注)。
  4. 在12孔板的每个孔中加入1ml生物素化混合物。将反应混合物室温孵育30分钟。
  5. 用PBS + 100 mM甘氨酸清洗细胞三次，以淬灭生物素化反应，并去除多余的生物素试剂和副产物。
  6. 细胞在室温冷藏甲醇中固定5分钟(或细胞在4% PFA(多聚甲醛)PBS中固定5分钟，在室温下PBS中洗涤三次，然后在0.1% Triton X-100中渗透10分钟，此步骤无需使用冷藏甲醇固定Sertoli细胞)。
  7. 在室温下用PBS洗涤三次。用阻断液(1% BSA in PBS, wt/vol)室温孵育30分钟。
  8. 吸出清除阻断液。加入~50 μL Streptavidin-FITC(Cat.No.GC19886)(含DAPI)，1:200稀释。室温孵育1小时。
  9. 在室温下用PBS洗涤三次。在黑暗中干燥，固定在载玻片上。

磷酸盐缓冲盐水(PBS)/CM: 10 mM NaH2PO4, 0.15 M NaCl, 1 mM CaCl2, pH 8.0, 22℃。
