Pseudo-UTP,100mM Sodium Solution is a modified uridine triphosphate nucleotide commonly used in the research and development of mRNA vaccines and therapeutic mRNAs. By incorporating into mRNA molecules in place of natural uridine, Pseudo-UTP can reduce the immunogenicity of mRNA while enhancing its stability and translational efficiency[1-2].

Pseudo-UTP合成SINEUP RNA

本实验方案根据研究内容整理,仅供参考,请根据实际需求进行调整。

1. RNA体外转录:在转录反应体系中,用Pseudo-UTP替代天然尿苷三磷酸(UTP),以掺入SINEUP RNA分子。修饰核苷酸需与未修饰核苷酸按特定比例混合(例如:m6A与Pseudo-UTP的组合需100%替代天然核苷酸)。通过部分替代GTP为m7G帽类似物,实现RNA 5'端加帽。

2.转录后处理:对体外转录的RNA进行纯化,并通过DNase I消化去除DNA模板。在纯化后,用大肠杆菌多聚腺苷酸聚合酶和ATP对RNA进行酶促加尾(约200nt),反应条件为37°C、30分钟。

3.功能验证:将修饰后的SINEUP RNA转染至293T/17细胞。转染48小时后,通过Western Blot分析目标蛋白表达水平,并通过qRT-PCR验证RNA稳定性及靶标mRNA表达量。

注意事项:

(1)Pseudo-UTP修饰可降低RNA免疫原性并提高核酸酶抗性,但需注意反转录效率可能受修饰影响。
