PD-1/PD-L1 inhibitor N11 is an inhibitor of the protein-protein interaction between programmed cell death protein 1 (PD-1) and PD-1 ligand (PD-L1; IC50 = 6.3 nM).[1] It increases T cell proliferation in a PD-L1-overexpressing CT26 colon cancer cell and T cell co-culture when used at a concentration of 20 nM. PD-1/PD-L1 inhibitor N11 (20 and 80 nM) promotes T cell infiltration into an in vitro PD-L1-overexpressing murine Lewis lung carcinoma tumor matrix. In vivo, PD-1/PD-L1 inhibitor N11 (20 mg/kg) reduces tumor growth and promotes CD8+ and CD4+ T cell tumor infiltration in a PD-L1-overexpressing murine Lewis lung carcinoma model.
