Nitrotetrazolium Blue chloride (NBT) is a dehydrogenase substrate that is used in western blots and immunohistochemical staining with the alkaline phosphatase (ALP) substrate 5-bromo-4-chloro-3-indolyl phosphate (BCIP) to produce a purple-blue precipitate[1]. Nitrotetrazolium Blue chloride can visualize the localization of target mRNAs at cellular resolution in embryos or tissues [2]. Nitrotetrazolium Blue chloride can react with reactive oxygen species (ROS) and superoxide anions (O2-) in cells or tissues to produce dark blue insoluble formazan compounds [3, 4]. Nitrotetrazolium Blue chloride can stain intracellular ALP and succinate dehydrogenase (SDH) [5, 6].

实验参考方法 Experimental Reference Method

本方案仅提供一个指导,请根据您的具体需要进行修改。

1. NBT和BCIP(GC39345)免疫印迹实验方案[1](来自文献,仅做参考)

(1)溶液配置:

NBT原液:将0.5g NBT溶解在10 mL 70% DMF水溶液中。

BCIP原液:将0.5g BCIP溶解在10 mL 70% DMF水溶液中。

底物缓冲液:100 mM 二乙醇胺缓冲液(pH 9.5),含有100mM NaCl和5 mM MgCl2

(2)制备NBT/BCIP溶液,将66 µL NBT原液添加到10 mL底物缓冲液中并充分混合。在溶液中加入33 μL的BCIP原液。1小时内使用NBT/BCIP溶液。

(3)将印迹膜放入合适的容器中并添加NBT/BCIP溶液。15 × 15cm2 的膜采用10mL溶液。在室温下搅拌并孵育,直至条带适当变暗,通常需要大约30 min。

(4)除去NBT/BCIP溶液,并用水冲洗印迹膜以终止反应。

2. NBT用于幼苗超氧化物积累的组织化学检测[2](来自文献,仅做参考)

(1)在琥珀色瓶中,将0.1g NBT溶于50mM磷酸钠缓冲液(pH 7.5)中,使体积至50ml,得到0.2% NBT染色液。

(2)将幼苗放入试管中,浸入NBT染色液中,用铝箔包裹管子,并在室温下过夜。

(3)倒掉染色液,将幼苗浸入无水乙醇中并在沸水浴中加热 10min(或必要时更长时间,间歇性摇晃)去除叶绿素。

(4)将幼苗转移到浸有60%甘油的纸巾上,在对比背景上拍摄染色的幼苗。

注意:确保所有溶液都是新鲜配制的,并且所有设备都是干净的,以避免污染。避免直接接触反应试剂。
