Lipid 35 is an ionizable cationic lipid (pKa = 7.6) that has been used in the generation of lipid nanoparticles (LNPs) for the delivery of mRNA in vitro and in vivo. LNPs containing lipid 35 selectively accumulate in the lungs, including in pulmonary epithelial, endothelial, myeloid, and dendritic cells, and spleen in mice. LNPs containing lipid 35 and encapsulating mRNA encoding Pseudomonas exotoxin A decrease the number of lung metastases and increase survival in a B16/F10.9 murine melanoma model.
