1. 试剂和材料准备
Isosulfan blue 1%（w/v）无菌生理盐水（0.9% NaCl）溶液，1mL无菌注射器，配27-30G针头，麻醉剂，荷瘤小鼠（BALB/c小鼠4T1乳腺癌模型）。
2. 实验步骤
（1）麻醉和体位调整
使用异氟烷或氯胺酮/赛拉嗪麻醉小鼠。将小鼠置于仰卧位并固定肢体。
（2）染料注射
将20-50µL Isosulfan blue装入注射器。在肿瘤部位附近皮下注射，在乳腺癌模型中通常注射至肿瘤周围或乳晕周围。注射部位：乳腺脂肪垫。轻轻按摩注射部位3-5分钟，以增强淋巴液的吸收。
（3）染料迁移和解剖
等待10-15分钟进行淋巴引流。根据肿瘤位置和预期的引流方式，在腹股沟或腋窝区域做切口。识别蓝色染色的淋巴管，并顺着它们找到蓝色哨兵淋巴结。切除并收集淋巴结进行组织学或分子分析。
注意事项
（1）注射量应较少，以防止组织肿胀和非特异性扩散。
（2）使用无菌技术避免感染。
（3）避免血管内注射；确保缓慢、可控的皮下给药。
（4）Isosulfan blue在极少数情况下可能引起过敏反应—请密切观察动物。
（5）使用适当的对照（未注射染料的小鼠）进行组织学比较。
