Hexa-His is an amino acid sequence tag composed of six consecutive histidine residues. Hexa-His achieves efficient protein purification based on immobilized metal ion affinity chromatography (IMAC) through the specific binding of the imidazole groups on histidine to metal ions. Hexa-His can be used in the purification of recombinant proteins, the study of protein-protein interactions, and research on protein-based supramolecular biopolymers[1-4].

Hexa-His标签亲和力测定[1]

本实验方案根据研究内容整理,仅供参考,请根据实际需求进行调整。

1.试剂准备:准备带有Hexa-His储备液,将DYE-tris-NTA荧光探针(BLUE-、GREEN-、RED-tris-NTA)溶解于PBST缓冲液(137mM NaCl,2.5mM KCl,10mM Na2HPO4,2mM KH2PO4,pH=7.4,0.05% Tween-20)中备用。

2. 亲和力测定:将带Hexa-His在PBST缓冲液中进行一系列稀释。向所有稀释管中加入固定浓度(50nM)的DYE-tris-NTA染料。将混合物在室温下避光孵育30分钟。将样品吸入专用毛细管,置于MicroScale Thermophoresis中,在合适的LED功率和中等MST功率下进行测量,通过结合曲线计算解离常数(Kd)。

注意事项:

(1)该方法对常见缓冲液添加剂(如BSA、还原剂TCEP/DTT)耐受性高,但应避免使用强螯合剂(如EDTA、EGTA)、离子型去垢剂(如SDS)和高浓度(>1mM)的竞争性物质(如咪唑、组氨酸)。
