实验参考方法 Experimental Reference Method

Z-Lys-SBzl . HCl测定补体因子D的酯解活性[1]

本实验方案根据研究内容整理,仅供参考,请根据实际需求进行调整。

1.试剂准备:Z-Lys-SBzl . HCl溶解于反应缓冲液中备用。配制0.10 M Hepes缓冲液(0.50M NaCl、9.8% DMSO)和4,4'-dithiodipyridine溶液。

2.反应体系建立:在比色杯中,建立1mL的最终反应体系。包含上述Hepes缓冲液、0.19mM Z-Lys-SBzl . HCl底物以及1μM的纯化人补体因子D(Factor D)。

3.酶活检测:将比色杯置于分光光度计中,在27°C条件下监测324nm波长处的吸光度变化。因子D水解Z-Lys-SBzl·HCl释放出的苯甲硫醇会立即与体系中的4,4’-二硫代二吡啶反应,生成4-硫代吡啶酮,该产物在324nm处的摩尔消光系数(ε)为19,800M⁻¹ cm⁻¹。通过吸光度的初始增长率计算酶促反应速率(v₀)。

注意事项:

(1)为验证反应完全,可在测定初始速率后,向体系中加入过量胰蛋白酶(如5μg),短时间内完全水解剩余底物,以精确计算底物的初始浓度。
