Hippuryl-Arg-OH is a fluorescent signal substrate used in enzyme activity assays for the identification and cleavage of peptide bonds containing arginine. Hippuryl-Arg-OH can be used for enzyme activity studies such as carboxypeptidase B and aminopeptidase, as well as related research in the field of drug development[1-4].

实验参考方法 Experimental Reference Method

Hippuryl-Arg-OH法测定血浆中TAFIa活性[1]

本实验方案根据研究内容整理,仅供参考,请根据实际需求进行调整。

1.试剂准备:试剂准备:将Hippuryl-Arg底物配制成20mM的工作液;将凝血酶(Thrombin)配制成5U/mL、血栓调节蛋白(Thrombomodulin)配制成7.5U/mL的工作液,两者混合形成凝血酶-血栓调节蛋白复合物;配制60mM的CaCl₂溶液;配制活化缓冲液(20mM HEPES,0.15M NaCl,5mM CaCl₂,pH=7.4);配制1M HCl溶液用于终止反应;配制1M NaOH溶液;配制1M磷酸盐缓冲液;配制溶于1,4-二氧六环的3%氰尿酰氯溶液。

2.TAFI活化:取15μL血浆样本,与12μL凝血酶-血栓调节蛋白复合物、8.5μL 60mM CaCl₂溶液以及24μL活化缓冲液混合。将混合液在室温下孵育10分钟,以将血浆中的TAFI活化为TAFIa。随后,加入20μL水蛭素(Hirudin)以终止活化反应。

3.TAFIa活性检测:向上一步得到的反应液中加入20μL 20mM的Hippuryl-Arg底物工作液,在室温下孵育10分钟。随后,加入20μL 1M HCl溶液以终止底物反应。然后,依次向反应体系中加入20μL 1M NaOH溶液和25μL 1M磷酸盐缓冲液。最后,加入60μL溶于1,4-二氧六环的3%氰尿酰氯溶液,并静置使其充分显色。

4.检测与计算:将完成显色的反应液在14,000g下离心2分钟。取上清液,使用光谱仪在405nm波长下测定其吸光度值。所有样品及标准曲线点均需用热灭活血清稀释至50%后,进行复孔测定。
