DDX5 蛋白, Human (His, Myc) 是重组的 DDX5 蛋白,由 E. coli 表达,带有 N-10*His, C-Myc 标签。

DDX5 是一种参与 pre-mRNA 剪接替代调控的 RNA 解旋酶,其活性依赖于 RBM4 并促进 tau 外显子 10 的包含。它能结合 tau pre-mRNA 外显子 10 下游的茎环结构区域,且单链 RNA 可显著刺激其 ATP 水解速率。该蛋白还参与转录调控(不依赖解旋酶活性),作为雄激素受体 AR(非 ESR1)的共激活因子,并与 DDX17 和 SRA1 RNA 协同激活 MYOD1 转录活性以促进骨骼肌分化。同时,它是 p53/TP53 的共激活因子,参与 DNA 损伤响应和凋亡,也是 RUNX2 的共激活因子以调控成骨细胞分化。此外,DDX5 能通过与 HDAC1 等组蛋白去乙酰化酶结合实现启动子特异性转录抑制,并作为大型 PER 复合物组分通过抑制 PER1 和 NR1D1 等节律基因的 3' 转录终止来调控昼夜节律。

Stability
Stored at -20°C for 2 years from date of receipt. After reconstitution, it is stable at 4°C for 1 week or -20°C for longer (with carrier protein). It is recommended to freeze aliquots at -20°C or -80°C for extended storage.
Biological Activity
Measured by its binding ability in a functional ELISA. Immobilized Human DDX5 at 2.5 μg/mL (100 μL/well) on the plate can bind Anti-DDX5 Antibody. The ED<sub>50</sub> for this effect is 36.69 ng/mL.
Formulation
Lyophilized from a 0.22 μm filtered solution of 10 mM Tris-HCl, 1 mM EDTA, 6% Trehalose, pH 8.0 or PBS, pH 7.4, 8% trehalose.
