Competence-Stimulating Peptide-2 (CSP-2) is a quorum-sensing signaling peptide produced by Streptococcus pneumoniae. Competence-Stimulating Peptide-2 activates bacterial competence by binding to compatible receptor ComD2 (EC50 of 50.7nM), thereby promoting the uptake of exogenous DNA and genetic transformation. Competence-Stimulating Peptide-2 can be used in studies related to quorum sensing, genetic transformation, and biofilm formation in Streptococcus pneumoniae, Streptococcus mitis, and Streptococcus oralis[1-4].

实验参考方法 Experimental Reference Method

Competence-Stimulating Peptide-2用于诱导肺炎链球菌自然感受态[1]
本实验方案根据研究内容整理,仅供参考,请根据实际需求进行调整。
1.试剂准备:将Competence-Stimulating Peptide-2配制成浓度为0.1mg/ml的工作液备用;准备感受态培养基(THY培养基,补充有0.5%牛血清白蛋白、0.2%葡萄糖和0.02% CaCl₂);制备供体菌的基因组DNA。
2.细菌预培养:将待诱导的肺炎链球菌菌株在感受态培养基中培养至对数生长中期(OD600≈0.2),温度为37℃,CO₂浓度为5%。
3.感受态诱导与转化程序:将预培养的菌液用新鲜感受态培养基按1:100稀释。取100μl稀释菌液,加入2μl浓度为0.1mg/ml的Competence-Stimulating Peptide-2和Competence-Stimulating Peptide-1工作液。将混合物置于37℃水浴中孵育12分钟。随后,向被肽刺激的菌液中加入供体DNA(500ng),并在30℃水浴中继续孵育20分钟,使外源DNA与细菌接触。
4.转化子筛选:将DNA-细菌混合液用新鲜感受态培养基稀释100倍,置于37℃水浴中复苏培养1.5小时。然后将菌液涂布于含有相应抗生素的选择性血琼脂平板上,于37℃、5% CO₂条件下培养过夜,以筛选转化子。
注意事项:
(1)Competence-Stimulating Peptide-2溶液应分装并储存在-20°C或更低温度,避免反复冻融。使用前应短暂离心,确保完全溶解和浓度准确。
