The cell-impermeable but visible light excitable Ca2+ indicator Calcium Green™-5N shows an increase in fluorescence emission intensity and a slight shift in wavelength upon binding Ca2+. Calcium Green™-5N is essentially nonfluorescent in the absence of Ca2+, and the low affinity indicator has a dissociation constant for Ca2+ in the absence of ∼14 µM Mg2+.
