C18 Ceramide-d7 (d18:1-d7/18:0) is a deuterated (d7) labeled C18 Ceramide isotope internal standard. Ceramide is a type of sphingolipid core molecule composed of a sphingosine (often d18:1) linked to a fatty acid via an amide bond[1]. Ceramides have been implicated in mediating or regulating numerous central cellular processes such as proliferation, differentiation and senescence, stress responses, apoptosis, and inflammation[2]. Harmful stimuli can activate ceramides, leading to a variety of pathological conditions[2]. C18 Ceramide is one type of ceramide containing 18-carbon fatty acids and is closely related to cardiovascular diseases[3]. C18 Ceramide-d7 is often used as an internal standard for LC-MS/MS to quantify C18 ceramide in plasma/serum/tissue/cells.

本方案来自文献,请根据您的具体需求修改。

1. 在甲醇中分别配置500pmol/μL的C18 Ceramide标准品溶液和0.05pmol/μL的C18 Ceramide-d7内标液。

2. 在10μL标准品溶液和血浆样品中各加20μL内标液,然后加入蛋白质沉淀(PPT)溶剂、乙酸乙酯:异丙醇(2:8,体积比)至每组终体积为600μL。混合10分钟,3000g离心10分钟后将上清液转移至96孔板中进行LC-MS/MS分析。

3. LC-MS/MS分析

流动相:(A)在LC-MS级水中加入0.1%甲酸的10mM乙酸铵溶液;(B)在体积比为4:3的乙腈和2-丙醇中加入0.1%甲酸的10mM乙酸铵溶液酸

进样量:5μL

流速:500μL/min

梯度变化:在0-0.5分钟,A/B为15/85%;0.5-4.0分钟,B比升至100%;4.0-5.0分钟,A/B回到15/85%

4. 计算每个样本中C18 Ceramide峰面积与C18 Ceramide-d7峰面积的比值,随后用校准曲线把峰面积比换算为血浆中C18 Ceramide的浓度。
