Description

Argireline (Acetyl hexapeptide-3) is a synthetic peptide that is patterned from the N-terminal end of the protein SNAP-25 [1]. Argireline inhibits vesicle docking by preventing formation of the ternary SNARE complex and by interfering in catecholamine release [2]. Argireline is used in cosmetic formulations to reduce facial muscle contractions, thus preventing the formation of expressive wrinkles, mainly on the forehead and around eyes[3].

In vitro, Argireline treatment for 48 hours significantly inhibited the viability of HEK-293 cells and IMR-32 cells, with IC50 values of 34.862μM and 68.458μM, respectively[4].

Preparation Method

HEK-293 cells were grown in DMEM medium with 10% (v/v) fetal bovine serum (FBS), 0.2mM glutamine, 100U/ml penicillin, and 100μg/ml streptomycin at 37°C in 5% CO2/atmosphere. Cells (1.5×105 cells/ml) were seeded in 96-well plates allowed to adhere in a 5% CO2 incubator at 37°C overnight. Cells were treated with different concentrations of Argireline (0.01, 0.1, 1, 10, and 100µM) for 48h. The viability was evaluated.

Applications

Argireline treatment significantly reduced the viability of HEK-293 cells in a dose-dependent manner.
