AIZin2 is a conditional labeling compound for zinc-binding proteins that contains a zinc-coordinating tridentate dipicolylamine group, an electrophile-reactive acyl imidazole moiety, and fluorescein diacetate.1 It is attached to proteins via an electrophilic attack on the acyl imidazole moiety when coordinated with protein-associated zinc. AIZin2 has been used to label zinc-binding proteins in HeLa cells for fluorescent detection and for LC-MS/MS proteomic experiments. It displays an emission at 559 nm upon excitation at 473 nm.
