2H,2H,3H,3H-Perfluorooctanoic acid (5:3 FTCA) is a perfluoroalkyl compound[1].

2H,2H,3H,3H-Perfluorooctanoic acid (5 mg; 48-144h) is a metabolite produced by the fungus Cunninghamella elegans during the biotransformation of 6:2 FTOH. Moreover, 2H,2H,3H,3H-Perfluorooctanoic acid, as the main metabolite, inhibits the activity of CYP enzymes, which are responsible for the biotransformation of 6:2 FTOH by Cunninghamella elegans[1].2H,2H,3H,3H-Perfluorooctanoic acid (10,000μg/L; 100 days) can be degraded by the aerobic bacterium Labrys portucalensis F11 (L. portucalensis F11) into PFHxA, PFPeA, PFBA, PFPrA, 5:3 fluorotelomer unsaturated carboxylic acid (5:3 FTUCA) and 7:2 fluorotelomer unsaturated carboxylic acid (7:2 FTUCA)[2].

L. portucalensis F11

Preparation Method

The biodegradation experiment of 2H,2H,3H,3H-Perfluorooctanoic acid by the L. portucalensis F11 strain was carried out under aerobic conditions in 250mL sealed flasks containing 50mL of minimal salts medium (MM) supplemented with 10,000μg/L of 2H,2H,3H,3H-Perfluorooctanoic acid. The cells of L. portucalensis F11 were inoculated at an optical density (OD) of approximately 0.05 at 600nm, and bacterial growth was monitored by measuring OD using a spectrophotometer.

All cultures were incubated at 30°C on a rotary shaker (130rpm). The experiments were performed in duplicate under sterile conditions. The abiotic control assays included sealed flasks containing MM supplemented with 10,000μg/L of 2H,2H,3H,3H-Perfluorooctanoic acid, without bacterial inoculation. Additionally, three flasks with the same concentration of methanol but without the addition of 2H,2H,3H,3H-Perfluorooctanoic acid were set up as controls for cell growth. Samples were taken for analysis at 0, 48, 100 and 194 days to assess microbial growth and the biodegradation of 2H,2H,3H,3H-Perfluorooctanoic acid. The purity of the cultures was evaluated by regularly plating on nutrient agar (NA) plates.

2H,2H,3H,3H-Perfluorooctanoic acid (10,000μg/L; 100 days) can be degraded by the aerobic bacterium Labrys portucalensis F11 (L. portucalensis F11) into PFHxA, PFPeA, PFBA, PFPrA, 5:3 fluorotelomer unsaturated carboxylic acid (5:3 FTUCA), and 7:2 fluorotelomer unsaturated carboxylic acid (7:2 FTUCA).
